Hepatocyte Regeneration

Hepatocyte regeneration is the process by which liver parenchymal cells restore tissue mass and function after injury or partial loss, making it a key model of tissue repair and developmental plasticity. In the mature liver, surviving hepatocytes commonly re-enter the cell cycle, replicate their DNA, and divide in response to cytokines, growth factors, and interactions with nonparenchymal cells; when proliferation is limited, progenitor or ductular cells may contribute. Studying these coordinated responses in developmental biology clarifies how cell fate, organ size, and tissue architecture are regulated. The findings inform research on liver disease, regenerative medicine, and therapies that support functional recovery.

Hepatocyte-specific Ablation in Zebrafish to Study Biliary-driven Liver Regeneration

To help assess the molecular mechanisms underlying zebrafish biliary-driven liver regeneration, we established a liver injury model in which the nitroreductase-expressing hepatocytes are genetically ablated upon metronidazole treatment. In this protocol, we describe how to adeptly manipulate, monitor and analyze hepatocyte ablation and biliary-driven liver regeneration.

Sustained fibrosis with deposition of excessive extracellular matrix proteins leads to cirrhosis. Alcohol abuse is one of the main causes of severe liver disease. We established an ethanol-induced zebrafish fibrotic liver model to study the mechanisms and strategies of promoting hepatocyte regeneration upon alcohol-induced injury.

We have described a protocol for performing partial hepatectomy (PHx) and cell transplantation via spleen in NOD.SCID (NOD.CB17-Prkdcscid/J) mice. In this protocol, an incision is made to expose and resect the left lobe of the liver followed by another incision for the intrasplenic transplantation of cells.
