Heart Metabolism

Heart metabolism is the network of biochemical processes that supplies energy and building materials for cardiac cells, supporting the heart’s contraction, growth, and adaptation. During development, cardiomyocytes adjust their use of substrates such as glucose and fatty acids as mitochondria mature and oxidative phosphorylation increasingly contributes to ATP production alongside glycolysis. These metabolic transitions influence cell proliferation, maturation, and structural remodeling, making heart metabolism an important framework for understanding normal cardiac development. Studying this process can clarify how metabolic disruption contributes to congenital heart defects, cardiomyopathies, and impaired regeneration, while informing strategies for developmental modeling and cardiac tissue engineering.

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

The goal of this protocol is to present a standard method to perform intravenous glucose tolerance tests (IVGTTs) to assess glycemic control in nonhuman primates and assess their metabolic status from healthy to dysmetabolic.

Normothermic Ex Situ Heart Perfusion in Working Mode: Assessment of Cardiac Function and Metabolism

Normothermic ex situ heart perfusion (ESHP), preserves the heart in a beating, semi-physiologic state. When performed in a working mode, ESHP provides the opportunity to perform sophisticated assessments of donor heart function and organ viability. Here, we describe our method for myocardial performance evaluation during ESHP.
