Liver Bud Progenitors

Liver bud progenitors are embryonic endoderm-derived cells that generate the liver and provide a model for understanding organ formation. During development, foregut endoderm cells respond to signals including fibroblast growth factors and bone morphogenetic proteins, forming the hepatic diverticulum and expanding into a liver bud; these progenitors then differentiate into hepatocytes and cholangiocytes. Studying their specification, migration, proliferation, and lineage commitment helps clarify liver development and congenital disease. Liver bud progenitors also support organoid construction, regenerative medicine, and disease modeling, offering experimental systems for testing how tissues self-organize and how damaged liver cells might be replaced.

Liver injuries are accompanied by progenitor cell expansion that represents a heterogeneous cell population. Novel classification of this cellular compartment allows for the distinguishing of multiple subsets. The method described here illustrates the flow cytometry analysis and high purity isolation of various subsets that can be used for further assays.

In Vitro Pancreas Organogenesis from Dispersed Mouse Embryonic Progenitors

The three-dimensional culture method described in this protocol recapitulates pancreas development from dispersed embryonic mouse pancreas progenitors, including their substantial expansion, differentiation and morphogenesis into a branched organ. This method is amenable to imaging, functional interference and manipulation of the niche.

The surface of the tongue is covered with various small bumps called papillae, which either distribute what has been ingested (filiform papillae) or contain the sensory taste (or gustatory) receptor cells (fungiform, circumvallate, and foliate papillae). Embedded within each taste-related papilla are the taste buds—clusters of 30 to 100 gustatory receptor cells. Gustatory receptor cells extend finger-like projections called gustatory hairs (or microvilli) into a region known as the taste pore.

Sustained fibrosis with deposition of excessive extracellular matrix proteins leads to cirrhosis. Alcohol abuse is one of the main causes of severe liver disease. We established an ethanol-induced zebrafish fibrotic liver model to study the mechanisms and strategies of promoting hepatocyte regeneration upon alcohol-induced injury.
