Optic Tectum Injury

Optic tectum injury is damage to the midbrain structure that receives and integrates visual information, providing a model for studying neural development and repair. In developing animals, injury can disrupt retinal axon guidance, synaptic connectivity, neuronal survival, and the organization of visual circuits, while subsequent responses may include inflammation, cell death, axon remodeling, and regeneration. Developmental biology research uses this injury paradigm to examine how neural tissues respond to damage, how developmental signals influence circuit reconstruction, and why some organisms recover visual function more effectively than others. These findings can clarify principles of neural plasticity and inform strategies for treating central nervous system injuries.

Stab Wound Injury Model of the Adult Optic Tectum Using Zebrafish and Medaka for the Comparative Analysis of Regenerative Capacity

A mechanical brain injury model in the adult zebrafish is described to investigate the molecular mechanisms regulating their high regenerative capacity. The method explains to create a stab wound injury in the optic tectum of multiple species of small fish to evaluate the regenerative responses using fluorescent immunostaining.

We describe the methods for fluorescent labeling of tangentially migrating cells by electroporation, and for time-lapse imaging of the labeled cell movement in a flat-mount culture in order to visualize migrating cell behavior in the developing chick optic tectum.
