Soil Microbial Analysis

Soil microbial analysis is the study of microorganisms in soil, including bacteria, fungi, archaea, and other microbial communities, to understand their roles in ecosystem function. Researchers assess microbial abundance, diversity, activity, and community composition using methods such as culture-based assays, microscopy, DNA sequencing, and measurements of respiration or enzyme activity. These analyses reveal how microbes drive decomposition, nutrient cycling, carbon storage, and soil formation, while showing how land use, pollution, climate, and agricultural practices alter soil health. In environmental sciences, the results support ecosystem monitoring, bioremediation, sustainable agriculture, and evaluation of environmental change.

2016

Phospholipid fatty acids provide information about the structure of soil microbial communities. We present methods for extraction from soil samples with a single-phase chloroform mixture, fractionation of extracted lipids using solid phase extraction columns, and methanolysis to produce fatty acid methyl esters, which are analyzed by capillary gas chromatography.

2012

We describe a method protocol for the GC-based analysis of the aldonitrile acetate derivatives of glucosamine and muramic acid extracted from soil. For elucidation of the chemical mechanism, we also present a strategy to confirm the structure of the derivative and the ion fragments formed upon electron ionization.

Soil Microbial Ecology

2026

Soil microbial ecology is defined by highly diverse, spatially structured communities that drive nutrient cycling, organic matter turnover, and overall ecosystem stability. Although a gram of soil can contain thousands of bacterial and archaeal taxa, the ecological processes they mediate are even more crucial for sustaining terrestrial life.Microhabitats and NichesSoil is a heterogeneous mixture of minerals, organic matter, water, and air. Microbes inhabit distinct microhabitats formed by...
