Dna Bait Strains

DNA bait strains are genetically engineered host cells that carry a defined DNA sequence, or bait, to test whether regulatory proteins bind that sequence. In a yeast one-hybrid assay, the bait is placed upstream of a reporter gene, while candidate proteins are fused to a transcriptional activation domain; binding recruits the activation domain and produces a measurable reporter signal under selective conditions. These strains help identify and characterize DNA-binding proteins, transcription factors, and sequence-specific regulatory interactions, supporting studies of gene regulation, promoter function, and genetic control of cellular processes.

The associated chromosome trap (ACT) assay is a novel unbiased method for identifying long-range DNA interactions. The characterization of long range DNA interactions will allow us to determine the relationship of nuclear architecture to gene expression in both normal physiology and in diseased states.
