Plasma Dna Extraction

Plasma DNA extraction is a laboratory method for isolating cell-free DNA (cfDNA) from blood plasma, enabling genetic analysis without collecting intact cells. After blood is separated to remove cellular components, cfDNA is captured on silica membranes or magnetic beads under high-salt conditions, washed to remove proteins and inhibitors, and eluted for downstream testing. In genetics, this workflow supports noninvasive prenatal testing, liquid biopsy, cancer monitoring, and detection of genetic or infectious disease markers. Careful processing is essential because contamination from genomic DNA released by damaged blood cells can distort DNA concentration and sequencing results.

We are describing a new method of isolating genomic DNA from whole blood collected for plasma/serology. After plasma collection, the compacted blood is usually discarded. Our novel method represents a significant improvement over existing methods and makes DNA and plasma available from a single collection, without requesting additional blood.

Source: Laboratories of Dr. Ian Pepper and Dr. Charles Gerba - The University of Arizona Demonstrating Author: Luisa Ikner Traditional methods of analysis for microbial communities within soils have usually involved either cultural assays utilizing dilution and plating methodology on selective and differential media or direct count assays. Direct counts offer information about the total number of bacteria present, but give no information about the number or diversity of populations present...

Circulating miRNA Extraction: A Method to Isolate miRNA from Plasma Samples Via Organic Extraction and Small RNA Enrichment

2023

This video describes the extraction of circulating microRNA from plasma samples of colorectal cancer patients using organic extraction followed by glass-fiber column enrichment. miRNAs are stable in biological fluids such as serum and plasma, which renders them ideal circulating biomarkers for cancer diagnosis, prognosis, and treatment decision-making and monitoring. This method will help identify new circulating biomarkers capable of predicting clinical outcomes in patients.
