Fresh Isolates

Fresh isolates are cells, microorganisms, or tissues obtained directly from a biological source and analyzed soon after collection, providing genetic material that has undergone minimal laboratory adaptation. In genetics, researchers typically recover individual specimens under sterile conditions, establish clonal or purified cultures when needed, and extract DNA or RNA for genotyping, sequencing, or other molecular assays. Because fresh isolates can retain variation present in the original population, they support studies of genetic diversity, mutation, inheritance, and genotype–phenotype relationships. They are also valuable for validating findings in established laboratory strains and for identifying naturally occurring variants relevant to evolution, disease biology, and biotechnology.

Changes in the intracellular calcium levels in the podocytes are one of the most important means to control the filtration function of glomeruli. Here we explain a high-throughput approach that allows detection of real-time calcium handling and single ion channels activity in the podocytes of the freshly isolated glomeruli.

This protocol describes a method for efficiently transfecting siRNA in freshly isolated human villous cytotrophoblasts using microporation and identifying DNA-protein complexes in these cells. Transfected cells can be monitored by Western blot and EMSA analyses during the 4-day culture time.
