Antimicrobial Testing

Antimicrobial testing is the systematic evaluation of how effectively drugs or other agents inhibit or kill microorganisms, helping guide infection treatment and monitor resistance. In laboratory assays, microbes are exposed to defined antimicrobial concentrations, and responses such as growth inhibition are measured to determine susceptibility, minimum inhibitory concentration, or minimum bactericidal concentration. Common approaches include disk diffusion, broth dilution, and time-kill testing, each revealing different aspects of antimicrobial activity. In immunology and infection research, these methods support antibiotic selection, investigation of host-pathogen interactions, evaluation of new therapies, and surveillance of emerging resistant strains.

Production and Testing of Antimicrobial Peptides and Their Mimics

This protocol describes a method for producing and evaluating the antimicrobial activity and cytotoxicity of the cationic peptide Mel4 and peptide mimic RK758. By integrating chemical synthesis, recombinant expression, and standardized functional assays, it provides a streamlined workflow for the production and testing of peptides and small-molecule mimics.

High Throughput, Real-time, Dual-readout Testing of Intracellular Antimicrobial Activity and Eukaryotic Cell Cytotoxicity

A high throughput, real-time assay was developed to simultaneously identify (1) eukaryotic cell-penetrant antimicrobials targeting an intracellular bacterial pathogen, and (2) assess eukaryotic cell cytotoxicity. A variation on the same technology was thereafter combined with digital dispensing technology to enable facile, high-resolution, dose-response, and two- and three-dimensional synergy studies.

Antimicrobial susceptibility testing of Mycobacterium tuberculosis is challenging but critical for patient care. This microtiter plate format offers testing of 12 antimycobacterial drugs and provides minimum inhibitory concentration (MIC) values, which will aid in appropriate treatment.

Application of the Intelligent High-Throughput Antimicrobial Sensitivity Testing/Phage Screening System and Lar Index of Antimicrobial Resistance

Here we introduce the principle, structure, and instruction of the intelligent high-throughput antimicrobial sensitivity testing/phage screening system. Its application is illustrated by using Salmonella isolated from poultry in Shandong, China, as an example. The Lar index is calculated, and its significance in evaluating antimicrobial resistance is discussed comprehensively.

Galleria mellonella as an Antimicrobial Screening Model

This study presents a standardized framework for optimizing G. mellonella infection models for use in preclinical antimicrobial assessment. The application of a G. mellonella model as part of a preclinical antimicrobial development pipeline could decrease the number of ineffective compounds progressing to clinical trials.
