Membrane-bound Il21

Membrane-bound IL-21 is a cell-associated form of interleukin-21, a cytokine that enables immune cells to communicate through direct contact and can shape antiviral and antibacterial responses. Displayed on the surface of activated immune cells or engineered feeder cells, it engages the IL-21 receptor on neighboring lymphocytes and activates JAK1/JAK3-STAT3 signaling, influencing proliferation, differentiation, and effector function. This mechanism supports interactions among T cells, B cells, and natural killer cells, including antibody production and cytotoxic activity. In immunology and infection research, membrane-bound IL-21 is used to study immune regulation and improve ex vivo expansion of therapeutic NK and T cells.

Here, we present a method to expand peripheral blood natural killer (PBNK), NK cells from liver tissues, and chimeric antigen receptor (CAR)-NK cells derived from peripheral blood mononuclear cells (PBMCs) or cord blood (CB). This protocol demonstrates the expansion of NK and CAR-NK cells using 221-mIL-21 feeder cells in addition to the optimized purity of expanded NK cells.

We describe here a technique that is now routinely used to isolate stably bound ribosome nascent chain complexes (RNCs). This technique takes advantage of the discovery that a 17 amino acid long SecM "arrest sequence" can halt translation elongation in a prokaryotic (E. coli) system, when inserted into (or fused to the C-terminus) of virtually any protein.
