Cell Engulfment

Cell engulfment is the process by which a cell surrounds and internalizes another cell, particle, or microorganism, making it important for tissue maintenance and innate immune defense. In phagocytosis, receptors recognize signals on the target, triggering actin cytoskeleton rearrangement that extends the plasma membrane around it and forms an intracellular phagosome; this compartment can then mature and fuse with lysosomes for degradation. In immunology and infection, cell engulfment enables macrophages and neutrophils to remove pathogens and dead cells, while its regulation helps explain host defense, inflammation, immune evasion, and potential therapeutic strategies.

Microglia are the resident immune cells of the central nervous system (CNS) with a high capacity to phagocytose or engulf material in their extracellular environment. Here, a broadly applicable, reliable, and highly quantitative assay for visualizing and measuring microglia-mediated engulfment of synaptic components is described.
