Complement Activity

Complement activity refers to the coordinated action of circulating and membrane-associated proteins that support immune defense by recognizing threats, promoting inflammation, and eliminating target cells. It begins through the classical, lectin, or alternative pathways, which converge on proteolytic cleavage of C3; resulting fragments such as C3b opsonize microbes, while downstream reactions can assemble the membrane attack complex. Measuring complement activity helps assess immune function, investigate recurrent infections, and characterize autoimmune or inflammatory disorders. In immunology and infection research, these responses clarify how innate and adaptive defenses interact and how pathogens evade or manipulate host immunity.

A Serum Bactericidal Assay for the Complement-Mediated Bactericidal Activity of Antibodies

This video demonstrates a technique to evaluate the complement-mediated bactericidal activity of antibodies present in serum. This approach combines isolated serum with pathogenic bacteria and exogenous complement proteins. Upon incubation, the bacteria are grown on an agar plate, and the produced colonies are quantified to measure the serum bactericidal titer.
