Cpg Odn 2006

CpG ODN 2006 is a synthetic oligodeoxynucleotide designed to mimic microbial DNA and activate innate immune signaling, making it a widely used tool in immunology and infection research. Its unmethylated CpG motifs are detected by Toll-like receptor 9 in endosomal compartments, triggering NF-κB and related pathways that promote B-cell activation, plasmacytoid dendritic-cell responses, and cytokine production. Researchers use this class B CpG agonist to model host responses to microbial DNA, assess immune adjuvants, and investigate how innate signals shape antibody production and antimicrobial defense.

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.

Synthesis of human monoclonal antibodies is the first step in studies aimed at unraveling the pathophysiological mechanisms of auto-antibody-mediated immune responses. We have developed a protocol to generate recombinant human immunoglobulin G (IgG) monoclonal antibodies from blood sorted B cells, including B-cell isolation, antibody cloning and in vitro synthesis.

Bisulfite amplicon sequencing (BSAS) is a method for quantifying cytosine methylation in targeted genomic regions of interest. This method uses bisulfite conversion paired with PCR amplification of target regions prior to next-generation sequencing to produce absolute quantitation of DNA methylation at a base-specific level.
