Dendritic Cell Migration

Dendritic cell migration is the directed movement of antigen-sampling immune cells from peripheral tissues to lymphoid organs, where it connects innate sensing with adaptive immunity. After detecting tissue damage or microbial signals, dendritic cells mature, increase expression of the chemokine receptor CCR7, and follow CCL19 and CCL21 gradients through lymphatic vessels to draining lymph nodes. There, they present processed antigens to T cells and provide the signals needed for immune activation or tolerance. Studying this process clarifies how infections initiate protective responses, how pathogens influence immune trafficking, and why disrupted migration can contribute to chronic inflammation, autoimmunity, or ineffective vaccination.

We describe a strategy to monitor maturation and migration of pulmonary dendritic cells in response to ovalbumin in the setting of ovalbumin induced allergic airway inflammation. This strategy can be modified to assess migration of pulmonary dendritic cells in settings of infection.
