Hsv Infection

HSV infection is a contagious viral disease caused by herpes simplex virus types 1 or 2, which establish lifelong infection after entering host cells. The virus attaches to and fuses with epithelial cells, delivers its double-stranded DNA genome to the nucleus, replicates, and then travels along sensory neurons to persist in a latent state; stress, illness, or immune changes can trigger reactivation and recurrent lesions. Studying HSV infection clarifies viral immune evasion, host inflammatory responses, and mechanisms of latency. These insights support diagnostic testing, antiviral treatment, and research into vaccines and therapies that could reduce transmission, recurrence, and complications in immunocompromised patients.

Source: Dembowski, J. A., Deluca, N. A. Purification of Viral DNA for the Identification of Associated Viral and Cellular Proteins. J. Vis. Exp. (2017)This video demonstrates the infection of stationary-phase mammalian cells with HSV-1 (Herpes Simplex Virus-1), followed by replication of the viral genome using host and viral replication machinery. A synthetic nucleoside analog is incorporated into newly synthesized viral DNA to label replicating genomes, which are subsequently isolated for...
