Igg3 Isotype

The IgG3 isotype is a subclass of immunoglobulin G antibodies that helps provide targeted defense against infectious agents. Its distinctive long hinge region and Fc domain support strong interactions with Fc gamma receptors and efficient activation of the classical complement pathway, promoting antibody-dependent cellular responses, opsonization, and pathogen clearance. IgG3 responses are especially relevant to immunity against protein antigens on viruses and bacteria, although their levels and functional activity can vary during infection. Studying this isotype helps researchers evaluate protective antibody responses, understand susceptibility to immunodeficiency, and inform vaccine development and diagnostic investigation.

This protocol describes tubulin purification from small/medium-scale sources such as cultured cells or single mouse brains, using polymerization and depolymerization cycles. In this paper, we describe a protocol to characterize T-dependent and T-independent immunoglobulin (Ig) isotype responses in mice using ELISA. This method used alone or in combination with flow cytometry will allow researchers to identify differences in B cell-mediated Ig isotype responses in mice following T-dependent and T-independent antigen immunization.

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.
