Lps Stimulation

LPS stimulation is an experimental method that activates immune cells with lipopolysaccharide, a major component of the outer membrane of Gram-negative bacteria, to model innate immune responses. LPS binds CD14 and the MD-2-TLR4 receptor complex, triggering MyD88- and TRIF-dependent signaling pathways that activate NF-κB and other transcription factors, leading to production of inflammatory cytokines and chemokines. In immunology and infection research, this approach helps characterize pathogen recognition, inflammatory signaling, macrophage and dendritic cell activation, and host responses to bacterial components. It also supports studies of sepsis, immune regulation, and candidate anti-inflammatory treatments.

Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.

Vagus nerve stimulation has proven to have a strong efficacy for decreasing peripheral inflammation. Here, we present a modified vagus nerve stimulation protocol that allows for further examinations of the cholinergic anti-inflammatory mechanisms in limited inflammatory responses.

We describe a simple immunoassay to measure the production of pro-inflammatory cytokines, such as IL-1 beta production, in patients presenting with autoinflammatory phenotypes. By activating cells in whole blood cultures with pathogen-associated molecular patterns, specifically with lipopolysaccharide, cytokine secretion can be conveniently evaluated in whole blood supernatants.
