Intra Hemocoelic Injection

Intrahemocoelic injection is a technique for delivering microorganisms, immune elicitors, drugs, or other test substances directly into the hemocoel, the body cavity of many invertebrates. A fine needle penetrates the cuticle and deposits the material into the hemolymph, allowing rapid distribution while bypassing external barriers and the gut. In immunology and infection research, this method enables controlled studies of host defense, pathogen virulence, antimicrobial responses, and survival. Researchers can standardize dose and exposure, then measure outcomes such as hemocyte activity, immune gene expression, microbial burden, or mortality, making the technique valuable for linking experimental challenge to invertebrate immune mechanisms.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.

Supramolecular hydrogelators based on ureido-pyrimidinones allow full control over the macroscopic gel properties and the sol–gel switching behavior using pH. Here, we present a protocol for formulating and injecting such a supramolecular hydrogelator via a catheter delivery system for local delivery directly in relevant areas in the pig heart.
