Intracellular Distribution Assessment

Intracellular distribution assessment is the analysis of where proteins, pathogens, nucleic acids, or other targets are located within cells, providing insight into cellular function and infection biology. It typically uses target-specific labeling followed by fluorescence imaging, quantitative microscopy, or cell fractionation to distinguish localization in compartments such as the cytoplasm, nucleus, endosomes, or lysosomes, while controls help separate true intracellular signal from surface-associated material. In immunology and infection research, these measurements reveal how microbes enter, traffic through, and persist in host cells, and how immune factors reach sites of action, supporting studies of pathogenesis, host defense, therapeutic delivery, and treatment response.

Near Infrared Optical Projection Tomography for Assessments of β-cell Mass Distribution in Diabetes Research

We describe the adaptation of optical projection tomography (OPT)1 to imaging in the near infrared spectrum, and the implementation of a number of computational tools. These protocols enable assessments of pancreatic β-cell mass (BCM) in larger specimens, increase the multichannel capacity of the technique and increase the quality of OPT data.
