Liquid Overlay

Liquid overlay is a laboratory method that places a defined liquid medium over cells, microorganisms, or biological samples to maintain contact and permit the exchange of soluble factors. During incubation, nutrients, signaling molecules, infectious particles, or antimicrobial compounds diffuse through the overlay, while changes in growth, viability, or infection provide measurable experimental outcomes. In immunology and infection research, liquid overlays can support controlled studies of host–pathogen interactions, microbial survival, and the effects of immune or therapeutic factors. The approach is useful for comparing conditions, recovering secreted products, and linking visible biological responses to specific components of the surrounding medium.

Plaque assays are the gold standard for viral quantification, utilizing entrapping overlays on host cellular monolayers to determine viral titers. While various semisolid overlays have traditionally been used, here we demonstrate plaque techniques comparing semisolid overlays to a novel liquid microcrystalline cellulose among several families of viruses.
