Liver Macrophages

Liver macrophages are innate immune cells that monitor the hepatic environment, remove cellular debris and pathogens, and help regulate inflammation and tissue repair. They include resident Kupffer cells, which reside in liver sinusoids, and monocyte-derived macrophages recruited during injury or infection; through pattern-recognition receptors, phagocytosis, and cytokine signaling, these cells detect danger and coordinate responses with hepatocytes, endothelial cells, and lymphocytes. In immunology and infection research, liver macrophages provide a model for studying pathogen clearance, immune tolerance, chronic inflammation, fibrosis, and host responses to systemic disease. Their functional plasticity also makes them important targets for therapies that modify hepatic and immune outcomes.

A Simple and Efficient Method to Isolate Macrophages from Mixed Primary Cultures of Adult Liver Cells

A novel method to obtain macrophages from primary culture of rat liver cells is described. This method utilizes the proliferation of macrophages in the culture, followed by shaking of culture flasks and purification by selective attachment to plastic dishes. This technique efficiently provides liver macrophages without complex equipment and skills.

Here, we study hepatotoxicity caused by lapatinib and neratinib through SASP-driven macrophage polarization, which offers a deeper mechanistic understanding beyond traditional toxicity tests. This approach demonstrates how senescent hepatocytes influence immune responses. This is particularly valuable for assessing long-term drug effects and discovering new therapeutic targets for managing drug-induced hepatotoxicity.

Macrophages play a central role in homeostasis and pathology in many tissues. The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice. These techniques allow determination of the role that polarized macrophages play in health and disease.
