Lmp Isolation Protocol

An LMP isolation protocol is a laboratory method for extracting latent membrane proteins, viral or cellular membrane-associated proteins that support studies of infection and immune recognition. The workflow typically disrupts cultured cells or infected material, solubilizes membrane proteins with an appropriate detergent, and separates the target fraction from insoluble debris while preserving protein structure for analysis. In immunology and infection research, isolated LMPs can support protein characterization, antigen-detection assays, antibody studies, and investigations of host-pathogen interactions. Standardized isolation improves reproducibility and helps researchers relate membrane-protein expression to viral persistence, cellular signaling, and immune responses.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.

Palatine tonsils are a rich source of B and T lymphocytes. Here we provide an easy, efficient and rapid protocol to isolate B and T lymphocytes from human palatine tonsils. The method described has been specifically adapted for studies of the viral etiology of tonsil inflammation known as tonsillitis.

Blood leukocytes and platelets can be used as a marker of overall bioenergetic health of an individual and so have the potential to monitor pathological processes and the impact of treatments. Here we describe a method to isolate and measure mitochondrial function and the oxidative burst in these cells.
