Lps Treatment

LPS treatment refers to exposing cells, tissues, or animal models to lipopolysaccharide, a bacterial outer-membrane component used to study innate immune activation and inflammation. LPS binds the CD14–MD-2–TLR4 receptor complex, triggering signaling pathways such as NF-κB and inducing cytokines, including tumor necrosis factor and interleukins. In immunology and infection research, this approach models aspects of bacterial challenge, endotoxemia, and sepsis while enabling controlled investigation of inflammatory responses. Researchers use LPS treatment to evaluate immune mechanisms, compare disease phenotypes, and assess potential anti-inflammatory or therapeutic interventions.

Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.

This video demonstrates an in vitro technique to study the impact of antimicrobial treatment on Pseudomonas aeruginosa (Pa) aggregates. Fluorescently-labeled Pa cells are grown as aggregates, mimicking the growth pattern during infection. Upon challenging the aggregates with a sublethal concentration of the antibiotic colistin, the real-time assessment of the bacteria-antibiotic interaction is performed via fluorescence microscopy.
