M1 Macrophage Activation

M1 macrophage activation is a pro-inflammatory immune state in which macrophages enhance antimicrobial defense and coordinate responses to infection. It is typically induced by signals such as interferon-γ and bacterial lipopolysaccharide, which engage intracellular signaling pathways, including NF-κB, to promote expression of inflammatory cytokines, chemokines, and microbicidal molecules. In immunology and infection research, M1 macrophages help restrict pathogens, recruit additional immune cells, and shape inflammatory responses. Excessive or prolonged activation, however, can contribute to tissue injury and chronic inflammation, making this process important for understanding host protection, disease mechanisms, and potential immune-modulating therapies.

Metabolic reprogramming is a characteristic and prerequisite for M1 and M2 macrophage polarization. This manuscript describes an assay for the measurement of fundamental parameters of glycolysis and mitochondrial function in mouse bone marrow-derived macrophages. This tool can be applied to investigate how particular factors affect the macrophage’s metabolism and phenotype.

Investigation of Macrophage Polarization Using Bone Marrow Derived Macrophages

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.

Following macrophage lysis, heat-inactivation of lysate, and centrifugation, intracellular caspases are collected for subsequent immunological analysis.

Macrophage phagocytic activity against cancer cells, specifically 4T1 mouse mammary carcinoma cells, was imaged in this study. The live cell coculture model was established and observed using a combination of fluorescent and differential interference contrast microscopy. This assessment was imaged using imaging software to develop multipoint time-lapse video.

Here, we present a detailed protocol for assessing the immunomodulatory potential of implant surfaces in vitro, aiming to improve the reliability and reproducibility of current protocols and promoting further research. Secretory cytokine profiles, mRNA expression, and cell surface markers were monitored using blood monocyte-derived macrophages to investigate macrophage polarization cultivated on titanium.
