Microcapillary Injection

Microcapillary injection is a microscale technique that delivers a defined volume of liquid into a cell, tissue, or small biological compartment, enabling controlled manipulation of otherwise inaccessible targets. A fine capillary, positioned with a micromanipulator, penetrates the target membrane or boundary, while applied pressure or displacement expels the selected material and limits delivery to the intended site. In immunology and infection research, this approach can introduce microbial components, immune reagents, or other experimental materials into individual cells for precise studies of host-pathogen interactions. It supports controlled perturbation, direct observation of cellular responses, and experiments requiring low-volume, spatially targeted delivery.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.

Supramolecular hydrogelators based on ureido-pyrimidinones allow full control over the macroscopic gel properties and the sol–gel switching behavior using pH. Here, we present a protocol for formulating and injecting such a supramolecular hydrogelator via a catheter delivery system for local delivery directly in relevant areas in the pig heart.
