Nickel Resin Chromatography

Nickel resin chromatography is a protein purification technique that isolates recombinant proteins engineered with a polyhistidine tag, supporting the production of defined reagents for immunology and infection research. The method uses immobilized nickel ions, commonly coordinated by nitrilotriacetic acid or related chelators, to bind histidine residues on the tag; contaminants are removed by washing, and imidazole or altered pH disrupts the interaction to elute the target protein. Researchers use this approach to purify microbial antigens, cytokines, antibodies, and other proteins for assays, structural studies, vaccine research, and host-pathogen investigations. Purity and protein stability depend on buffer composition and chromatography conditions.
