Physical Map Development

Physical map development is the process of determining the actual order and spacing of DNA sequences, genes, or other landmarks along a chromosome or genome, providing a structural framework for genetic analysis. Researchers construct these maps by identifying sequence-tagged sites, restriction landmarks, or overlapping DNA clones and using shared markers or clone overlaps to assemble regions into ordered contigs, with distances expressed in base pairs. In immunology and infection research, physical maps organize host immune-response loci and pathogen genomes, support positional cloning and genome sequencing, and help relate genomic structure to virulence, antigen variation, or disease susceptibility. These maps also aid genome assembly validation and comparative studies.
