Thymocyte Overlay

Thymocyte overlay is an ex vivo assay that places thymocytes on a prepared stromal-cell layer or tissue section to examine how developing T cells interact with their microenvironment. During the assay, thymocytes settle onto the substrate under defined culture conditions, allowing receptor-mediated adhesion, contact-dependent signaling, and, when relevant, migration or retention to be assessed by microscopy or cell recovery. In immunology and infection research, this approach helps characterize thymic selection, stromal support, and changes in thymocyte behavior after controlled immune perturbations. It provides a practical bridge between molecular studies and the organization of T-cell development in the thymus.

In this video, we describe a method to identify the small-molecule kinase inhibitors that modulate the apoptosis of self-reactive CD4+CD8+ double-positive immature thymocytes. Apoptosis is induced in the double-positive thymocytes by activating them with anti-CD3- and anti-CD28-coated magnetic beads; this is followed by a small-molecule inhibitor treatment and flow cytometry analysis to detect if the inhibitors modulate the apoptotic marker expression.

Plaque assays are the gold standard for viral quantification, utilizing entrapping overlays on host cellular monolayers to determine viral titers. While various semisolid overlays have traditionally been used, here we demonstrate plaque techniques comparing semisolid overlays to a novel liquid microcrystalline cellulose among several families of viruses.
