Biliary Pancreatitis

Biliary pancreatitis is acute inflammation of the pancreas caused by gallstones or biliary sludge obstructing the pancreaticobiliary outflow, making it a common and clinically important form of acute pancreatitis. Obstruction near the ampulla of Vater disrupts digestive secretions and promotes premature activation of pancreatic enzymes within the gland, leading to tissue injury and inflammation. Diagnosis combines characteristic abdominal pain with elevated pancreatic enzymes and imaging findings, while ultrasound or other imaging can identify gallstones and bile duct obstruction. Management focuses on supportive care, treating complications, relieving persistent obstruction when indicated, and preventing recurrence through gallbladder removal.

To help assess the molecular mechanisms underlying zebrafish biliary-driven liver regeneration, we established a liver injury model in which the nitroreductase-expressing hepatocytes are genetically ablated upon metronidazole treatment. In this protocol, we describe how to adeptly manipulate, monitor and analyze hepatocyte ablation and biliary-driven liver regeneration.
