Crypt Plating

Crypt plating is a cell culture technique that isolates intestinal crypts, the tissue structures containing stem and progenitor cells, and grows them under controlled laboratory conditions. Researchers release crypts from intestinal tissue through mechanical disruption and enzymatic digestion, then embed them in a supportive extracellular matrix and provide defined growth factors that promote three-dimensional organoid formation. In medicine, crypt plating enables investigation of epithelial regeneration, intestinal disease, host responses, and patient-specific tissue behavior. The resulting organoids support drug screening, toxicity studies, and research into personalized therapies while reducing reliance on animal models.

In this protocol, we describe how to generate murine primary epithelial colon monolayers directly from intestinal crypts. We provide experimental approaches to generate confluent monolayers on permeable filters, confluent monolayers for scratch wound healing and biochemical studies, and sparse and confluent monolayers for immunofluorescence analysis.

When working with media and reagents used to culture microorganisms, aseptic technique must be practiced to ensure contamination is minimized. A variety of plating methods are routinely used to isolate, propagate, or enumerate bacteria and phage, all of which incorporate procedures that maintain the sterility of experimental materials.
