Elderly Cancer Patients

Elderly cancer patients are older adults diagnosed with malignancy whose care requires attention to both tumor biology and age-related health changes. Aging can alter organ function, drug metabolism, immune responses, mobility, cognition, and nutritional status, while comorbidities and frailty may influence tolerance of surgery, chemotherapy, radiation, or immunotherapy. Geriatric assessment evaluates these factors alongside cancer stage and treatment goals, helping clinicians tailor therapy, manage symptoms, prevent complications, and support independence. This integrated approach strengthens shared decision-making and may improve quality of life while balancing disease control with the risks and benefits of treatment.

This manuscript details a method used to generate prostate cancer patient derived xenografts (PDXs) from circulating tumor cells (CTCs). The generation of PDX models from CTCs provides an alternative experimental model to study prostate cancer; the most commonly diagnosed tumor and a frequent cause of death from cancer in men.

Zebrafish Patient-Derived Xenograft (zPDX): A Cancer Model

This video describes the technique to create a zebrafish patient derived xenograft model. In the sample protocol, we assess the efficiency of the drugs Gemcitabine and Navitoclax on the pancreatic tumor xenograft model.

Patient Derived Cell Culture and Isolation of CD133+ Putative Cancer Stem Cells from Melanoma

This article describes the preparation of freshly obtained melanoma tissue into primary cell cultures, and how to remove contaminations of erythrocytes and fibroblasts from the tumor cells. Finally, we describe how CD133+ putative melanoma stem cells are sorted from the CD133- bulk using Magnetic Activated Cell Sorting (MACS).

Utilizing patient-derived tumors in a subcutaneous preclinical model is an excellent way to study the efficacy of novel therapies, predictive biomarker discovery, and drug resistant pathways. This model, in the drug development process, is essential in determining the fate of many novel anti-cancer therapies prior to clinical investigation.
