Global M6a Levels

Global m6A levels refer to the overall abundance of N6-methyladenosine, a chemical modification found throughout cellular RNA, and provide a broad measure of RNA methylation status. These levels reflect the balance between methyltransferase complexes that add m6A, demethylases that remove it, and reader proteins that recognize the modification to influence RNA stability, processing, translation, and decay. In medicine, measuring global m6A levels can help characterize altered RNA regulation in disease, support comparisons between healthy and pathological tissues, and complement transcript-specific analyses. As a result, this measure contributes to research on epitranscriptomic biomarkers and potential therapeutic targets.

Dot Blot Assay for Detecting Global N6-Methyladenosine RNA Modification Levels

2026

Here, we present a protocol to semi-quantitatively assess global m⁶A levels using dot blot. Total RNA is extracted, denatured, spotted on a nylon membrane, probed with anti-m⁶A antibody, and visualized by chemiluminescence. Signal intensity, quantified by ImageJ grayscale analysis, reflects relative methylation abundance, providing a reproducible workflow for research.

We describe a technique for analysis of global RNA synthesis in hypoxia using imaging. Click-chemistry labeling of RNA has not previously been performed under hypoxia and allows visualization of global RNA changes at the single cell level. This approach complements the existing averaged RNA techniques, allowing direct visualization of cell-to-cell changes in global RNA synthesis.
