Glutamine Uptake

Glutamine uptake is the transport of the amino acid glutamine from extracellular fluid into cells, where it supports cellular metabolism, biosynthesis, and nitrogen balance. Specialized membrane transporters, including sodium-dependent systems such as ASCT2 and SNAT proteins, regulate entry according to transporter activity, ion gradients, and extracellular glutamine availability. In medicine, measuring or modifying glutamine uptake helps investigators study tumor metabolism, immune-cell activation, tissue repair, and metabolic adaptation during disease. Because altered uptake can reflect changes in cell growth and nutrient demand, it is also relevant to identifying therapeutic vulnerabilities and evaluating treatments that target cancer or cellular metabolism.

This article will demonstrate how to monitor glutamine dynamics in live cells using FRET. Genetically encoded sensors allow real-time monitoring of biological molecules at a subcellular resolution. Experimental design, technical details of the experimental settings, and considerations for post-experimental analyses will be discussed for genetically encoded glutamine sensors.

Radiolabeled Amino Acid Uptake Assay to Quantify Cellular Uptake of Amino Acids

Here we present a step-by-step protocol of the long-length electrostatic repulsion-hydrophilic interaction chromatography-tandem mass spectrometry (LERLIC-MS/MS) method. This is a novel methodology that enables for the first time quantification and characterization of the glutamine and asparagine deamidation isoforms by shotgun proteomics.
