Intact Pericardium Model

The intact pericardium model is an experimental preparation that preserves the pericardial sac surrounding the heart, enabling cardiovascular physiology to be studied under near-natural mechanical conditions. Because the relatively noncompliant pericardium encloses the heart, changes in cardiac volume and pressure can alter pericardial pressure, ventricular filling, and interactions between the ventricles. This model helps investigators examine cardiac mechanics, hemodynamics, and the effects of conditions such as pericardial effusion, tamponade, and constriction. It also supports evaluation of how interventions influence heart function when external constraint and ventricular coupling remain intact, improving the physiological relevance of preclinical cardiovascular research.

This study demonstrates the feasibility and safety of developing an autologous pulmonary valve for implantation at the native pulmonary valve position by using a self-expandable Nitinol stent in an adult sheep model. This is a step toward developing transcatheter pulmonary valve replacement for patients with right ventricular outflow tract dysfunction.

Single-cell electroporation (SCE) is a specialized technique allowing delivery of DNA or other macromolecules into individual cells within intact tissue, including in vivo preparations. Here we detail the procedure for SCE of a fluorescent dye or plasmid DNA into neurons within the intact brain of the Xenopus laevis tadpole.
