Rodent Retina Imaging

Rodent retina imaging is the use of optical methods to visualize and measure retinal structure, blood vessels, and disease-related changes in laboratory rodents, providing a noninvasive window into ocular and systemic health. Depending on the instrument, fundus photography records reflected light from the retinal surface, optical coherence tomography uses low-coherence interferometry to generate cross-sectional tissue images, and fluorescence imaging tracks labeled cells or vascular leakage. These approaches enable longitudinal assessment of retinal degeneration, inflammation, vascular injury, and therapeutic response in models of human disease. Repeated imaging can reduce animal use while linking anatomical changes with functional and molecular findings in biomedical research.

Source: Wang, Z., et al. Transpupillary Two-Photon In Vivo Imaging of the Mouse Retina. J. Vis. Exp. (2021).This video demonstrates a technique for in vivo retinal imaging using two-photon microscopy. Anesthetized mice are prepared for imaging by securing their heads, Applying eye lubricant, and placing a coverslip over the eye. Retinal ganglion cells (RGCs) expressing fluorescent proteins are first visualized using epifluorescence for alignment. Finally, two-photon imaging is employed,...

Creating experimental retinal detachments with a reproducible and sustained height of detachment, and without subretinal hemorrhage, is important for studying the pathophysiology of photoreceptor cell loss in retinal disease and evaluating potential therapeutic interventions. Here, we report such a method in detail.

2010

This protocol describes three Drosophila preparations: 1) adult brain dissection, 2) adult retina dissection and 3) developing eye disc- brain complexes dissection. Emphasis is laid on special preparation techniques and conditions for live imaging, although all preparations can be used for fixed tissue immunohistochemistry.
