Tissue Normalization

Tissue normalization is the process of standardizing tissue samples, measurements, or images so that biological differences can be distinguished from variation caused by collection, preparation, or analysis. It works by applying consistent sampling and processing conditions, then adjusting measurements against reference features or baseline values to reduce technical and structural variability. In medicine, tissue normalization supports reliable comparison across patients, experiments, and time points, strengthening histopathology, imaging, biomarker analysis, and treatment evaluation. By improving data consistency, it helps researchers identify disease-associated changes and supports the development of more reproducible diagnostic and therapeutic approaches.

Cancer Associated Fibroblasts (CAFs) facilitate tumor initiation, growth and progression through signaling that promotes proliferation, angiogenesis, and inflammation. Here we describe a method to isolate pure populations of normal fibroblasts and CAFs from fresh mouse and human tissues by cell sorting, using PDGFRα as a surface marker.
