Toggle Clamp Sealing

Toggle clamp sealing is a mechanical method for closing and securing a lid, panel, or access port by using a toggle clamp to maintain compressive force around a seal. As the handle moves through an over-center position, the linkage converts manual input into clamping pressure and locks the assembly in place, compressing a gasket or sealing surface to limit fluid, gas, or particulate leakage. In medicine and biomedical engineering, this approach supports secure closure of equipment housings, sterilization containers, laboratory devices, and fluid-handling systems. Reliable sealing helps maintain containment, protect sterile conditions, simplify repeated access, and improve the consistency of device operation.

Patch Clamp Electrophysiology

Neuron cell membranes are populated with ion channels that control the movement of charge into and out of the cell, thereby regulating neuron firing. One extremely useful technique for investigating the biophysical properties of these channels is called patch clamp recording. In this method, neuroscientists place a polished glass micropipette against a cell and apply suction to form a high resistance seal. This process isolates a small “patch” of membrane that contains one or more ion channels.

Key procedures to optimize the sealing process and achieve real-time monitoring of the metal-to-glass seal (MTGS) structure are described in detail. The embedded fiber Bragg grating (FBG) sensor is designed to achieve online monitoring of temperature and high-level residual stress in the MTGS with simultaneous environmental pressure monitoring.

Described here is a procedure for obtaining long stretches of current recording from one ion channel with the cell-attached patch-clamp technique. This method allows for observing, in real time, the pattern of open-close channel conformations that underlie the biological signal. These data inform about channel properties in undisturbed biological membranes.
