Axon Trafficking Assessment

Axon trafficking assessment is the analysis of how molecular cargo moves through axons, long neuronal projections that depend on precisely directed transport to maintain signaling and function. The process relies primarily on microtubule tracks and motor proteins, including kinesin and dynein, which carry vesicles, proteins, organelles, and other materials between the cell body and axon terminal. Researchers assess cargo distribution, movement, direction, and transport dynamics using approaches such as live-cell imaging and quantitative analysis. These measurements help reveal how neurons maintain polarized architecture and identify trafficking defects associated with neurodevelopmental disorders, neurodegeneration, and impaired neuronal communication.

Use of pHluorin to Assess the Dynamics of Axon Guidance Receptors in Cell Culture and in the Chick Embryo

We describe here the use of a pH-sensitive green fluorescent protein variant, pHluorin, to study the spatio-temporal dynamics of axon guidance receptors trafficking at the cell surface. The pHluorin-tagged receptor is expressed both in cell culture and in vivo, using electroporation of the chick embryo.

Analysis of SCAP N-glycosylation and Trafficking in Human Cells

We describe a modified method for membrane fraction isolation from human cells and sample preparation for the detection of SCAP N-glycosylation and total protein by using western blot. We further introduce a GFP-labeling method to monitor SCAP trafficking using confocal microscopy. This protocol can be used in regular biology laboratories.

While the transport of cell surface proteins is relatively easily studied, visualizing the trafficking of intracellular proteins is much more difficult. Here, we use constructs incorporating photoactivatable GFP and demonstrate a method to accurately follow the amyloid precursor protein from the Golgi apparatus to down-stream compartments and follow its clearance.
