Cm-h2dcfda Staining

CM-H2DCFDA staining is a fluorescence-based method for detecting intracellular reactive oxygen species (ROS), making it useful for assessing oxidative stress in cells, including neurons. The cell-permeable probe enters cells, where intracellular esterases remove its acetate groups; subsequent oxidation by ROS converts the nonfluorescent compound into fluorescent dichlorofluorescein, allowing signal intensity to reflect relative ROS levels under defined conditions. In neuroscience, this assay helps evaluate oxidative damage associated with neuronal activity, toxic exposures, disease models, and protective treatments. Combined with appropriate controls and quantitative imaging or plate-based measurements, it supports comparisons of redox changes across experimental groups, although fluorescence is generally interpreted as a relative indicator rather than a direct measurement of a single ROS species.
