Electrophysiological Methods

Electrophysiological methods are techniques that measure the electrical activity of cells, tissues, or neural circuits, providing direct insight into how the nervous system communicates. In neuroscience, electrodes detect voltage changes or current flow, while approaches such as patch-clamp recording measure membrane potentials and ion-channel activity in individual neurons, and extracellular recordings capture signals from populations of cells. These methods reveal action potentials, synaptic transmission, network dynamics, and changes in excitability. Their applications range from investigating basic mechanisms of neural signaling to studying sensory processing, neurological disorders, and the effects of drugs or stimulation on brain function.

Electrophysiological Methods for Recording Synaptic Potentials from the NMJ of Drosophila Larvae

Here we describe electrophysiological methods for measuring synaptic transmission at the neuromuscular junction of Drosophila larva. Evoked release is initiated artificially by stimulating the motor neuron axons, and transmission through the NMJ can be measured by the postsynaptic response evoked in the muscle.

Different electrode coatings affect neural recording performance through changes to electrochemical, chemical and mechanical properties. Comparison of electrodes in vitro is relatively simple, however comparison of in vivo response is typically complicated by variations in electrode/neuron distance and between animals. This article provides a robust method to compare neural recording electrodes.
