Gustatory Neurons

Gustatory neurons are sensory nerve cells that transmit information about taste, linking chemical stimuli in the mouth to neural representations in the brain. When molecules dissolved in saliva activate taste receptor cells within taste buds, those cells change their electrical activity and release signals to connected nerve fibers, which carry information through cranial nerves to brain regions involved in perception and feeding behavior. Studying gustatory neurons helps explain how the nervous system distinguishes basic taste qualities and integrates taste with other sensory cues. This research supports investigations of appetite, nutrition, taste disorders, and therapies for altered taste perception.

New Methods to Study Gustatory Coding

We present three new methods to study gustatory coding. Using a simple animal, the moth Manduca sexta (Manduca), we describe a dissection protocol, the use of extracellular tetrodes to record the activity of multiple gustatory receptor neurons, and a system for delivering and monitoring precisely timed pulses of tastants.

In this protocol, we describe two strategies that simultaneously suppress two genes (double gene knockdown) in honey bees. Then we present how to use the proboscis extension response (PER) assay to study the effect of double gene knockdown on honey bee gustatory perception.

Detecting Physiological Responses from Sensory Neurons in Drosophila

This video demonstrates a method for detecting sensory neuron responses in Drosophila using ligand binding and fluorescence imaging. Ligands bind to neuron receptors, causing calcium influx. The calcium binds to indicator complexes, leading to fluorescence emission, which confirms the neuronal response to the ligand.

The forelegs and proboscis of Drosophila contain a rich repertoire of gustatory sensory neurons. Here, we present a method using calcium imaging to measure physiological responses from sensory neurons in the foreleg and proboscis of live flies upon exogenous application of a gustatory pheromone.
