Insulin Tolerance Test

The insulin tolerance test is a physiological method that evaluates how the body responds to insulin-induced hypoglycemia, providing insight into insulin sensitivity and neuroendocrine function. Under controlled conditions, insulin lowers blood glucose, while serial measurements of glucose and hormones such as ACTH, cortisol, or corticosterone track metabolic recovery and activation of counterregulatory pathways. In neuroscience, the test is used to study hypothalamic regulation of energy balance, stress responses, and the hypothalamic-pituitary-adrenal axis, as well as to assess endocrine function after disruptions affecting the brain or pituitary. Because hypoglycemia can be severe, the procedure requires careful monitoring and standardized recovery measures.

Conserved insulin signaling pathways found in the fruit fly Drosophila melanogaster make this organism a potential tool for modeling metabolic disorders including type II diabetes. To this end, it is critical to establish physiological assays to effectively measure systemic insulin action in peripheral glucose disposal in the adult fly.

Begin with a culture of Alishewanella, a Gram-negative, salt-tolerant bacterium.Next, streak the culture onto agar plates containing increasing concentrations of sodium chloride to simulate varying levels of saline stress.Overlay each plate with a layer of unsolidified medium to prevent oxygen diffusion and maintain the culture in an oxygen-limited environment.Seal and incubate the plates at an optimum temperature to support bacterial growth.During incubation, the bacterium exhibits varying...
