Microglial Content Assessment

Microglial content assessment is the quantitative evaluation of microglia, the resident immune cells of the central nervous system, within neural tissue or experimental samples. Researchers typically label microglia with cell-type markers such as Iba1 or TMEM119, then measure cell density, tissue coverage, morphology, or activation-associated markers using microscopy, image analysis, or flow cytometry. These measurements help characterize neuroinflammatory responses, microglial proliferation, migration, and changes linked to brain injury, neurodegenerative disease, infection, or treatment. By providing objective cellular data, microglial content assessment connects tissue-level immune activity with neuronal function and disease mechanisms in neuroscience research.

Assessing Retinal Microglial Phagocytic Function In Vivo Using a Flow Cytometry-based Assay

2016

Microglial phagocytosis is critical for the maintenance of tissue homeostasis and inadequate phagocytic function has been implicated in pathology. However, assessing microglia function in vivo is technically challenging. We have developed a simple but robust technique for precisely monitoring and quantifying the phagocytic potential of microglia in a physiological setting.

Assessing Microglial Phagocytosis of Myelin Debris in vitro Under Repeated Magnetic Stimulation

The present protocol has been devised to assess the effect of repetitive magnetic stimulation on microglia's ability to phagocytose myelin debris. An in vitro microglia and myelin debris co-culture system has been established to do so.

Take adhered murine microglial cells in a multiwell plate.Add polyacrylic acid-coated magnetic nanoparticles (MNPs) to the test well, leaving another well untreated as a control.Incubate the cells to allow MNP uptake via endocytosis, enabling their internalization.Wash with buffer to remove unbound MNPs.Add an enzyme solution to detach the cells.Transfer the detached cells to tubes and centrifuge to pellet them.Discard the supernatant, resuspend the pellet in basal media, and transfer the...
