Notch Cleavage Analysis

Notch cleavage analysis is the study of proteolytic processing of the Notch receptor, a key signaling event that regulates cell fate decisions in neural development and adult nervous-system function. When a neighboring cell presents a Notch ligand, receptor activation triggers sequential cleavage, including gamma-secretase-dependent release of the Notch intracellular domain, which enters the nucleus and alters transcription. Researchers use cleavage analysis to track receptor activation, distinguish signaling states, and examine how changes in ligand interaction, protease activity, or receptor processing affect neural progenitor maintenance, differentiation, and disease-related pathways. The approach supports mechanistic studies of cell-to-cell communication in neuroscience.

RNA polymerase II synthesizes a precursor RNA that extends beyond the 3' end of the mature mRNA. We have successfully generated two substrate-specific γ-secretase assays. Both cell-based assays presented here are designed to quantify γ-secretase enzymatic activities via the output of firefly luciferase reporters.
