Parkinsonian Zebrafish

Parkinsonian zebrafish are zebrafish models that reproduce key features of Parkinson’s disease, providing a practical system for studying dopaminergic neurodegeneration and motor dysfunction. Researchers generate these models through genetic manipulation or exposure to neurotoxic compounds, which can reduce dopamine-producing neurons and produce measurable changes in swimming, movement, and behavior. Their small size, rapid development, optical accessibility, and suitability for high-throughput experiments support investigations of disease mechanisms, cellular pathways, and therapeutic targets. Parkinsonian zebrafish also enable efficient screening of candidate drugs and evaluation of neuroprotective strategies, complementing mammalian models while helping clarify how neuronal damage affects whole-organism behavior.

Here, we present a protocol for the isolation of increasingly insoluble/aggregated alpha-synuclein (α-syn) from post-mortem human brain tissue. Through the utilization of buffers with increasing detergent strength and high-speed ultracentrifugation techniques, the variable properties of α-syn aggregation in diseased and non-diseased tissue can be examined.

In vivo Electroporation of Morpholinos into the Adult Zebrafish Retina

A method to conditionally knockdown a target protein’s expression in the adult zebrafish retina is described, which involves intravitreally injecting antisense morpholinos and electroporating them into the retina. The resulting protein is knocked down for several days, which allows testing the protein’s role in the regenerating or intact retina.
