Phototaxis Verification Test

The Phototaxis Verification Test is a behavioral assay that determines whether an organism moves toward or away from light, providing a measurable readout of sensory and neural function. During the test, controlled illumination stimulates photoreceptors, which convert photons into neural signals that modify locomotor circuits; researchers then quantify direction, preference, response latency, or movement speed. In neuroscience, this assay helps evaluate visual processing, sensory-motor integration, and the integrity of neural pathways. Comparisons across genetic backgrounds, developmental stages, drug treatments, or injury models can reveal how specific biological changes affect light-guided behavior and support investigations of nervous system function.

Source: Weber, N. et. al., Natural Transformation, Protein Expression, and Cryoconservation of the Filamentous Cyanobacterium Phormidium lacuna. J. Vis. Exp. (2022)This video demonstrates a focused LED-based assay for studying phototaxis in filamentous cyanobacteria. The bacteria migrate toward the light source, forming a dense accumulation at the illuminated center, which is visualized and quantified through pixel intensity analysis of the captured image.

Self-Evaluation: Self-Enhancement and Self-Verification

2020

Social psychologists have documented that feeling good about ourselves and maintaining positive self-esteem is a powerful motivator of human behavior (Tavris & Aronson, 2008). In the United States, members of the predominant culture typically think very highly of themselves and view themselves as good people who are above average on many desirable traits (Ehrlinger, Gilovich, & Ross, 2005). Often, our behavior, attitudes, and beliefs are affected when we experience a threat to our...

Strategies of Self-Presentation II: Self-Verification

2025

Self-verification is a fundamental psychological drive wherein individuals seek affirmation of their self-concept from others, striving for consistency between their internal self-view and external perceptions. This drive operates even when the self-concept is negative, influencing interpersonal behavior and feedback preferences in complex and often counterintuitive ways. Unlike the self-enhancement motive, which seeks positive evaluations, self-verification prioritizes coherence and...

Failure of Cleaning Verification in Pharmaceutical Industry Due to Uncleanliness of Stainless Steel Surface

The lack of a well-defined procedure that consistently cleaned coupon surfaces was identified as the major contributor to low and variable recoveries in cleaning verification. This manuscript describes the correct protocol of cleaning stainless steel coupons.

Light Spot-Based Assay for Analysis of Drosophila Larval Phototaxis

2019

This protocol introduces a light-spot assay to investigate Drosophila larval phototactic behavior. In this assay, a light spot is generated as light stimulation, and the process of larval light avoidance is recorded by an infrared light-based imaging system.
