Subculture Protocol

A subculture protocol is a standardized procedure for transferring proliferating cells from an established culture into fresh growth conditions, allowing the population to be maintained, expanded, and studied over time. In neuroscience, the process typically involves assessing cell health and confluence, gently dissociating or detaching cells, and reseeding them at a controlled density in sterile, nutrient-containing medium that supports neuronal or glial growth. Consistent subculturing helps preserve viable cultures, reduce overcrowding, and produce reproducible cell populations for experiments involving neurodevelopment, cellular signaling, disease modeling, and drug response. Careful timing and handling are essential because culture conditions can influence cell phenotype and experimental outcomes.

Source: Anwar, Z., et al. Enrichment and Detection of Clostridium perfringens Toxinotypes in Retail Food Samples. J. Vis. Exp. (2019).In this video, sulfite-reducing bacteria are cultured in a sulfite-containing agar medium, where they grow anaerobically and reduce sulfite to hydrogen sulfide, forming black precipitates with iron ions. Black colonies are isolated from the agar depth using aseptic techniques and transferred to fresh medium for continued anaerobic incubation.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.
