Carbon Linker Variants

Carbon linker variants are alternative carbon-based spacers that connect functional groups or pharmacophores within a drug molecule, making them important tools in medicinal chemistry and pharmacology. By changing linker length, branching, flexibility, or ring structure, researchers can control the three-dimensional arrangement of binding groups and modify properties such as target affinity, selectivity, lipophilicity, metabolic stability, and membrane permeability. Comparing these variants helps establish structure–activity relationships and identify compounds with improved pharmacokinetic and pharmacodynamic profiles. This strategy supports lead optimization, target-probe design, and the development of safer, more effective therapeutic candidates.

We describe a set of assays to analyze expression levels of H1 linker histones. mRNA of individual H1 genes are quantitatively measured by random primer based reverse transcription followed by real-time PCR, whereas protein quantification of H1 histones is achieved by HPLC analysis.

Identification of Sleeping Beauty Transposon Insertions in Solid Tumors using Linker-mediated PCR

A method of identifying unknown drivers of carcinogenesis using an unbiased approach is described. The method uses the Sleeping Beauty transposon as a random mutagen directed to specific tissues. Genomic mapping of transposon insertions that drive tumor formation identifies novel oncogenes and tumor suppressor...

Cytoskeletal Linker Proteins - Plakins

2023

Plakins are large proteins with binding domains for microtubules, microfilaments, intermediate filaments, and membrane-associated protein complexes at cell junctions. Plakin functions are evolutionarily conserved and are primarily involved in organizing the different components of the cytoskeleton by crosslinking them to each other and connecting them to the cell-matrix and cell adhesion complexes. They are also known to interact with signal transducers, serve as scaffolds for signaling...

Histone Variants at the Centromere

2020

Histone variants are the histone proteins with structural and sequence variations. These variants may be regarded as “mutant” forms that replace their canonical histone counterparts in the nucleosomes. Specific post-translational modifications on the histone variants enable further chromatin complexity and regulate tissue-specific gene expression. The most common histone variants are from histone H2A, H2B, and linker histone H1 families. However, several variants of histone H3 variants are also...

Generation of Influenza Virus Escape Variants against Neutralizing Antibodies

2025

This video demonstrates a technique for generating escape variants of neutralizing influenza virus monoclonal antibodies. Antibody-neutralized virus suspension is injected into embryonated chicken eggs to propagate escape variants selectively. This results in a virus population predominantly of escape variants, confirmed via a Hemagglutination assay.
