Intracellular Calcium Concentration

Intracellular calcium concentration is the amount of free Ca²⁺ within a cell, a tightly regulated signal that controls processes ranging from muscle contraction and secretion to gene expression and cell survival. Cells adjust calcium levels through release from intracellular stores such as the endoplasmic reticulum, influx through voltage-gated or receptor-operated channels, and removal by pumps and exchangers that restore resting conditions. In pharmacology, measuring these calcium dynamics helps reveal how drugs modify ion channels, receptors, and signaling pathways, supporting research on cardiovascular, neurological, muscular, and secretory disorders. Calcium imaging and related assays can also identify drug efficacy, selectivity, and potential cellular toxicity.

Monitoring Changes in the Intracellular Calcium Concentration and Synaptic Efficacy in the Mollusc Aplysia

We demonstrate how changes in the intracellular free calcium concentration and synaptic efficacy can be simultaneously monitored in a ganglion preparation of Aplysia. We image intracellular calcium using a fluorescent dye, Calcium Orange, and induce and monitor synaptic transmission with sharp (intracellular) electrodes.
