神経変性におけるニューロンとグリアの相互作用は、ツールや方法が不十分なため、よく理解されていません。ここでは、ヒト多能性幹細胞から誘導されたニューロン、希突起膠細胞前駆細胞、および希突起膠細胞を取得するための最適化されたプロトコルについて説明し、アルツハイマー病における細胞種特異的な寄与を理解する上でのこれらの方法の価値の例を提供します。

要約

アルツハイマー病(AD)やその他の神経変性疾患では、希突起膠障害は一般的な初期の病理学的特徴ですが、特に脳の灰白質において、それが疾患の発症と進行にどのように寄与するかはほとんど不明のままです。希突起膠細胞系譜細胞の機能不全は、髄鞘形成の欠損および希突起膠細胞前駆細胞(OPC)の自己複製障害によって特徴付けられる。これらの2つの欠陥は、病理学の蓄積に沿ったニューロンと希突起膠細胞の間の相互作用の混乱によって少なくとも部分的に引き起こされます。OPCは、CNSの発生中に有髄性希突起膠細胞を生じさせる。成熟した脳皮質では、OPCは主要な増殖細胞(全脳細胞の~5%を占める)であり、神経活動依存的に新しいミエリン形成を制御します。このようなニューロンから希突起膠細胞へのコミュニケーションは、適切なツールがないため、特にADなどの神経変性状態の状況では、かなり研究されていません。近年、私たちのグループなどは、ヒト多能性幹細胞から機能的なニューロンと希突起膠細胞を個別に生成するために、現在利用可能なプロトコルを改善するための大きな進歩を遂げました。本稿では、ニューロン-希突起膠細胞結合をモデル化するための共培養システムの確立など、最適化された手順について説明します。私たちの実例となる結果は、OPC /希突起膠細胞が脳アミロイドーシスとシナプスの完全性に予想外の寄与をしていることを示唆しており、AD研究におけるこの方法論の有用性を強調しています。この還元主義的アプローチは、脳内の固有の複雑さから特定のヘテロ細胞相互作用を分析するための強力なツールです。ここで説明するプロトコルは、神経変性の病因における希突起膠原欠陥に関する将来の研究を促進することが期待されます。

概要

希突起膠細胞前駆細胞(OPC)、有髄性希突起膠細胞、およびその間の移行型を含む希突起膠細胞系譜細胞は、ヒト脳細胞の主要なグループを構成しています1神経発達と老化を通して中枢神経系の適切な操作と維持のための多くの重要な機能に積極的に関与しています2,3,4.希突起膠細胞は、白質のニューロン活動伝達を促進し、軸索の健康をサポートするミエリンを産生することでよく知られています、OPCは髄鞘形成がほとんどない灰白質に豊富(~5%)存在し、学習行動と記憶形成を制御する活動依存性シグナル伝達機能を実行します5,6,7,8 .アルツハイマー病(AD)やその他の加齢に伴う神経変性状態の病因における希突起膠細胞がどのように機能し、機能不全になるかは十分に研究されていません9。適切なモデルシステムの不十分さと、実験の道を前進させるための一般的な知識の不足が、このギャップの主な理由です。

プロトコル

1. ヒト多能性幹細胞からのヒトニューロン誘導

  1. レンチウイルス調製(~5日、前述の詳細なプロトコル16)
    1. 各T75フラスコに~100万個のHEK293T細胞をプレートし、トランスフェクションを行う際にそれらを~40%コンフルエントにさせる。テトラサイクリン誘導性Ngn2およびピューロマイシン耐性遺伝子(PuroR;同じTetOプロモーター制御下)、rtTA、および3つのヘルパープラスミドpRSV-REV、pMDLg/pRRE、およびVSV-G(レンチウイルスベクターDNA12 μgおよびヘルパープラスミドDNAのそれぞれ6 μg)を発現するプラスミドをトランスフェクトします。レンチウイルス調製物ごとに少なくとも3つのフラスコを準備します。トランスフェクションには、製造元の指示に従ってPEIを使用してください。16時間後にメディアを交換し、廃棄します。
    2. 毎日培地を採取して放出されたウイルス粒子を収穫し、3日間新鮮な培地と交換します。精製のためにウイルス粒子を含む収集された培地をプールします。ウイルスを0.22 μmのフィルターでろ過し、49,000 x g で90分間遠心分離します。ペレットを適切な容量のPBSグルコース(~150 μL)に再懸濁します。
  2. ニューロン誘導(~5日)

結果

ヒト多能性幹細胞からのヒト誘導神経細胞の直接生成
出発ヒト多能性幹細胞がiNまたはiOPC / iOLの生成を成功させるために高度な多能性を示すことは非常に重要です。したがって、本稿に記載されている誘導プロトコルのいずれかを開始する前に、Oct4やSOX2などの特定のマーカーについて細胞を染色する必要があります(図1A)。ヒトH1細胞を用いて、Zhangらによって以前に発表されたプロトコルに従って、いくつかの変更を加えて誘導された興奮性前脳ニューロンを得た(図1C)12161718ここでは、2日目のiNを、グリアまたは線維芽細胞などのフィーダー層がない状態で、マトリックス溶液(材料の表を参照)

ディスカッション

希突起膠細胞系譜細胞は、シナプス構造を安定化させ、髄鞘形成による塩状シグナル伝導を促進するための物理的および代謝的サポートに加えて、ニューロンとの迅速かつ動的なクロストークを介してニューロン活動パターンを形成することができます5,6,7AD病理学では、オリゴデンドログリア応答は当初、炎症と酸化ストレスに続発するだけと見なされていましたが、現在では、ミエリンの完全性の低下がAβ凝集とタウ過剰リン酸化の出現前の初期の病原性イベントであると主張する有望な証拠があります9。さらに、OPCの自己複製による髄鞘形成の修復は、ニューロン活動に大きく依存するプロセスであるAD38で特に脆弱です。したがって、健康なニューロン希突起膠細胞シグナル伝達をサポートするメカニズムを理解することは、新しい治療標的を特定する絶好の機会となります。

Co-culturing of iNs and iOPCs
This protocol is optimized specifically for co-culturing iNs and iOPCs and allow our real-time monitoring of the inter-cellular communications between these two cell types along the course of neural development. The ideal plating densities for both cell types need to be decided with a series of cell number titration to achieve proper differentiation (Figure 3A). After 4 weeks in co-cultures, the iOPCs are expected to be adequately differentiated into OLs that are positive for specific markers such as MBP and extend processes to ensheath axons (Figure 3B). The co-culture system can robustly boost up the number of synapses, indicating that the iOPCs provide a neuronal support through physical contacts or release of trophic factors (Figure 3C). We can maintain the co-cultures in acceptable health condition for up to 6 weeks and observe that the synapse number and other neuronal attributes plateau around the fifth week. Of note, astrocytes and microglia are not present in our preparations and their absence can be documented by checking the expression of specific markers (Figure 3D).
